CJC-1295
GHRH Analog + GH Secretagogue Combination
Also: No DAC) + Ipamorelin Blen
Reviewed
Overview
This combination pairs CJC-1295 without Drug Affinity Complex (also known as Mod GRF 1-29) with Ipamorelin to create a synergistic growth hormone releasing stack. CJC-1295 (No DAC) is a modified form of GHRH (growth hormone releasing hormone) consisting of the first 29 amino acids with four amino acid substitutions that increase stability.
The combination works through complementary mechanisms: CJC-1295 stimulates the GHRH receptor pathway while Ipamorelin activates the ghrelin/GHS receptor. When used together, they produce a greater GH pulse than either peptide alone, while retaining ipamorelin's receptor selectivity.
Mechanism of action
The combination leverages two distinct but synergistic pathways:
CJC-1295 (GHRH Analog) Mechanism Binds to and activates the GHRH receptor on pituitary somatotrophs, stimulating GH synthesis and release. The modified amino acids (positions 2, 8, 15, 27) prevent DPP-IV degradation, extending half-life from minutes to approximately 30 minutes.
Ipamorelin (GHS) Mechanism Activates the ghrelin receptor independently of the GHRH pathway. This amplifies the GH pulse initiated by CJC-1295 while maintaining selectivity (no cortisol, prolactin, or appetite effects).
Synergistic Effects The two pathways converge on the somatotroph cell, where their combined signaling produces a GH release greater than the sum of individual effects. This mimics the natural interplay between GHRH and ghrelin in physiological GH regulation.
Pulsatile Release The combination maintains natural pulsatile GH patterns rather than sustained elevation, preserving receptor sensitivity and more closely approximating physiological GH secretion.
